ULTRAMICROTOMY TECHNICIAN

Position and Tasks to be developed.
The candidate will be responsible for servicing ISABIAL’s Electron Microscopy (EM) Platform.
This platform will allow ISABIAL researchers to have the latest technologies for high-resolution 3D structure-preserved electron microscopy (EM) imaging by high-pressure freezing and correlate them with fluorescence microscopy (CLEM) images.
The main functions and tasks to be performed will be, among others.
– Technical advice to users on the appropriate preparation techniques in each type of sample for fluorescence microscopy or electron microscopy.
– Performing the processes prior to the serial cutting of the samples.
– High-pressure freezing of samples (HPF).
– Cutting of material for optical microscopies, such as the kerosene microtome, vibratome, cryostat and ultramicrotome.
– Staining with dyes.
Mounting of samples.
– Ultrathin slices for electron microscopy with samples embedded in resin or high pressure (HPF).
– Staining and contrasting of grids.
– Immunolabeling service of paraffin sections, vibratome, cryostat or ultrathin (preinclusion, postinclusion or Tokoyasu).
– Operation of the Axio Observer 7 Fluorescence microscope with Apotome3 (Zeiss).
– Operation of GeminiSEM 460 FE-SEM microscope (Zeiss) and Atlas 5 software (with Array Tomography software, ZEN Connect and ZEN CAT.
Correlative Array Tomography).
– Maintenance of microscopes and equipment of the ISABIAL microscopy and histology platform.
– Purchase and storage of reagents from suppliers.
– Waste management of the ISABIAL Microscopy Platform.
– Implementation of new optical, electron, immunolabeling and CLEM microscopy protocols.
– Train and educate future personnel of other services that collaborate with the platform.
– Management and preparation of budgets and invoices to users and customers of the platform.
– Quality control and maintenance of Leica processing equipment and Zeiss microscopes.
– Help search for new funding sources for the platform and training courses.
